I’ve got 2TB free space on my 4TB drive. One partition is 2TB, encrypted with LUKS. Using Gnome Disks, Is it possible to resize this, or expand it to 4TB and take up the whole drive? Or do I need to backup and create a 4TB partition?

I think, that the better way is backup and recreate partition.
It’s not that difficult, but make sure to backup your data first,
The more difficult part is editing the partition table. Cryptsetup and most filesystems i.e btrfs do support resizing even online (btrfs, xfs , ext4).
gnome disks? no idea, probably yes. Best boot a current live image. You could also use a gparted live image instead.

I’m following up on your issue, as I have a similar one (see post here).
I’m considering recreating a new partition and importing my LUKS-encrypted system files and my home files in it, but I don’t understand how I can restore those files.
Do you have any hint on how to restore system files? Or at least: the apps I installed, extensions and my Fedora settings?
